Baricitinib is a selective JAK1 and JAK2 inhibitor. It appears as off-white or white crystalline powder, odorless and tasteless, and is mainly indicated for the treatment of moderate to severe active rheumatoid arthritis (RA) in adult patients.Approved by the European Union in February 2017 as the first JAK inhibitor for RA treatment, the drug can be administered either as monotherapy or in combination with methotrexate. In June 2018, it obtained US FDA approval for RA patients with inadequate response to TNF inhibitors.In June 2022, the US FDA expanded its approval for the treatment of severe alopecia areata in adults. Besides, it has also been approved in the EU and Japan for moderate-to-severe atopic dermatitis (AD).
